摘要
目的:明确缺血预处理的刺激量与其心肌细胞保护效果的关系。方法:以Wistar大鼠为模型,开胸安置缺血再灌注装置。将48只大鼠分为4组,每组各12只:A组持续阻断冠状动脉1小时,再灌注2小时;B组缺血5分钟,再灌注5分钟,阻断冠状动脉1小时,再灌注2小时;C组缺血5分钟,再灌注5分钟,重复3次,阻断冠状动脉1小时,再灌注2小时;D组缺血10分钟,再灌注5分钟,阻断冠状动脉1小时,再灌注2小时。检测血浆肌酸激酶浓度与心脏切片氯化硝基四氮唑蓝染色,判断心肌梗死范围;磁带记录心电,分析心律失常。结果:缺血预处理对缺血心肌有明显的保护作用;增加刺激量(缺血预处理的时间与次数),没有增加保护效果。结论:心肌细胞的缺血预处理保护可能依靠阈值触发,无论刺激强弱,一旦启动则是充分完全地发挥保护作用。
? Objective:To determine the relationship between the duration or strength of Ischemic preconditioning and its protective effect aganist the cardiac infarction and arrhythmia in rats. Methods:The snare for occlusion and reperfusion had been put in to the main coronary artery in 48 Wistar rats which were divided into 4 groups.Each group received the different strength or duration of stimulation in ischemic preconditioning.The infarct size was estimated by NBT staining and plasma concentration of creatine kinase (CK).The cardiac arrhythmia recorded by tape recorder was analysed with the computer. Results:The protective effect of ischemic preconditioning was evident.As the stimulation of ischemic preconditioning became stronger,no apparent change took place in the protective effect,the concentration of CK was higher(p<005). Conclusion:The protective effect aganist the cardiac infarction and arrhythmia in rats may be triggered by the threshold of preconditioning.The stimulation of ischemic preconditioning above the threshold is not beneficial to myocardial protection in rats.
