摘要
目的:探讨建立成年大鼠心肌细胞钙超载模型的方法。方法:Langendorff灌流,分离钙耐受成年大鼠心肌细胞,采用不同浓度钙离子导入剂-伊屋诺霉素(ionomycin)作用心肌细胞1小时,用荧光分光光度计测定心肌细胞内的游离钙浓度。结果:1.分离的活性心肌细胞比率约70~80%,杆状、横纹清晰。2.实验组1(ionomycin0.5μmol/L)心肌细胞[Ca^2+]1明显高于对照组(282.07±25.19 vs 115±21.21nmol/L,P〈0.05);实验组2(ionomycin 1.0μmol/L)心肌细胞[Ca^2+]1(376.35±29.44)显著高于正常对照组(P〈0.05)。结论:通过本方法可以建立成年大鼠心肌细胞钙超载模型。
Objective: To develop the calcium overload model with adult rat ventricular cardiomyoeytes.Methods: Calcium tolerant cardiomyocytes were isolated by Langendorff perfusion. Then, cardiomyocytes were exposed to ionomycin for 1 hour, and cytosolic calcium was measured with spectrofluorometer. Results: Viability of freshly isolated adult rat ventricular cardiomyocytes, which were rod- shaped with clear cross- striations, was 60% - 80% . Cytosolic calcium of cardiomyocytes stimulated by 0.5μmol/L or 1.0μmol/L ionomycin increased significantly by 145 % ( P 〈 0.05, vs control) and 226 % ( P 〈 0.05, vs control) respectively, Conclusion: Calcium overload model of adult rat ventricular cardiomyocytes can be built by ionomycin.
