摘要
采用高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法,以C18(4.6 mm×250 mm,5μm)为色谱柱,水-乙腈梯度洗脱,流速1.0 mL·min^-1,PL-ELSD 2100为检测器,外标法定量,以桔梗皂苷D为标准品建立了以8种主要成分为质量控制指标的含量测定方法,并对该法进行方法学考察.结果表明:桔梗皂苷D含量在4.168~20.84μg范围与峰面积呈线性关系,检测的精密度RSD为0.958%,稳定性RSD为1.33%,重复性RSD为2.98%,添加回收率平均值为95.60%,RSD为1.91%.该方法灵敏、准确,重复性好,回收率高,操作简便,可用于桔梗饮片的质量控制.
In this paper, HPLC-ELSD method is established to determine the content of eight main components by using platycodin as standard. Liehrospher C18 column (4.6 mm × 250 mm, 5 μm) is used and gradient eluted by water-acetonitrile is chosen at speed 1.0 mL · min^-1. PL-ELSD 2100 detector is used for qualitative and quantitative analysis in external standard method. Results indicate that in the range of 4. 168~20. 84 μg platycodin D has a good linearity with precision RSD is 0. 958%, stability RSD is 1.33%, reproducibility RSD is 2.98%, average recovery rate of 95.60%, RSD is 1.91%. This method is sensitive, accurate, reproducible and easy to be operated with high recovery rate.
