摘要
用磷脂酰胆碱、神经节苷酯、胆固醇以45∶10∶45的克分比浓度制备出大的单层脂质体,在其水相溶液中分别包被0.5,1,2,4mg的碱性磷酸酶(AKP),经或不经TritonX-100处理后,与AKP的底物作用,于400nm处测OD值。结果显示,当AKP为1mg/ml时,效果最理想(0.784±0.056和0.175±0.027)。该脂质体充入氮气后,于4℃中可保存3个月。用过碘酸钠法分别偶联5,10,15mg/ml的抗AFP于脂质体上,发现随着抗AFP浓度的增加,偶联的抗AFP的比例升高,而结合抗AFP的百分比却有下降趋势。
Large unilamellar liposomes were prepared from phosphatidylcholine, ganglioside, and cholesterol in a 45, 10 and 45 molaration and contained 0.5, 1,2, 4mg/ml alkaline phosphatase (AKP) respectively in their aqueous phase . The realease of entrapped AKP was recorded on OD400nm by the addition of the substrate with liposomes either treated or not treated by Triton X100. The results were the best (0.784±0.056 and 0.175±0.027) when the concentration of AKP was 1mg/ml. Thee stability could last 3 months when the liposomes were stored at 4℃ under atmosphere of nitrogen. The results of 5,10 and 15mg AFP coupled seperately to liposomes treated by the periodate demonstrated that the greater the concentration of antiAFP was the high the ratio of the antiAFPcoupled to liposome to phosphatide was , and the lower the percentage of bound antiAFP was.
