摘要
建立了套式引物聚合酶链式反应用于Ⅰ型艾滋病毒pol基因的检测,并比较了常规单对引物PCR和套式引物PCR的敏感性。结果表明,当模板DNA分子的数量低于1000个时,常规单对引物PCR反应不能检出,而套式引物PCR反应则可检出低至10个分子的模板DNA。提示采用套式引物可以大大提高聚合酶链式反应的敏感性。
The nested primer polymerase chain reaction (PCR) was established for the detection of HIV1 pol gene.The sensitivity of nested PCR was compared with that of routine PCR.The results showed that PCR with nested primers can detect even 10 molecules of template DNA,but the routine PCR can not detect the template DNA molecules less than 1 000.The results suggested that PCR with nested primers could greatly promote the sensitivity of polymerase chain reaction.
