摘要
目的:建立血浆中不对称二甲基精氨酸浓度的反相高效液相色谱测定法(RP-HPLC)。方法:采用C_(18)柱(150 mm×4.6 mm,5μm),以0.05 mmol·L^(-1)磷酸二氢钾(pH3.0):甲醇(72:28)为流动相,流速1.0 ml·min^(-1),荧光检测:激发波长338 nm,发射波长425 nm,邻苯二甲醛(OPA)柱前衍生,柱温35℃。结果:不对称二甲基精氨酸在40~200 ng·ml^(-1)范围内有良好线性关系,高中低浓度平均回收率均大于90%,日内、日间RSD小于8%。结论:本方法准确、灵敏,适用于血浆不对称二甲基精氨酸浓度检测。
To establish a pre-column derivatization HPLC method for determination of Asymmetric dimethylarginine (ADMA) in plasma. Method: OPA was employed as a derivative, A C18 column( 150 mm × 4.6 mm ,5 μm)was used as a stationary phase, mobile phase was composed of Methanol- 0.05mol·L^-1 potassium dihydrogen phosphate (28: 72), flow rate was 1.0ml·min^-1, excitation wavelength was 338nm, emission wavelength was 425 nm and column temperature was at 35℃. Result: The linear concentration range of Asymmetric dimethylarginine was 40 - 200μg. ml^-1( r = 0. 998 6 ) , the recovery rates of high, medium and low concentration were larger than 90% , with RSD less than 8% , The drug-containing plasma was stable at least in 3 weeks under -80℃. Conclusion : The elaborated method is simple and practicable, and suitable for monitoring drug level of Asymmetric dimethylarginine in plasma.
