摘要
meso-四(N-甲基-3-吡啶基)卟啉和钴的配合物(CoTMPyP)可以作为辣根过氧化物酶的模拟酶来催化高香草酸和H_2O_2的反应,生成具有强荧光的高香草酸的二聚体。用CoTMPyP催化,可以测定3.0×10^(-7)—2.0×10^(-6)mol/L的H_2O_2,通过和葡萄糖氧化酶反应的偶联,可以测定0.20—4.0μgmL^(-1)的葡萄糖。对血清中的葡萄糖含量进行了测定。
The catalytic behaviour of a mimetic enzyme of peroxidase, cobalt tetrakis (N-methylpyridinumyl) porphyrin (CoTMPyP), is demonstrated in a fluorescence reaction of homovanillic acid (HVA) with hydrogen peroxide. The assays for hydrogen peroxide and glucose are linear in the range of 3.0 × 10-7 - 2. 0 × 10-5 mol/1 of hydrogen peroxide and 0.20-4.00 μg/ml of glucose, respectively. The recommended method has been applied to determine glucose in human serum, satisfactory results were obtained.
