摘要
目的观察基因重组转化生长因子α┐绿脓杆菌外毒素融合蛋白(TGFα┐PE40)对表皮生长因子受体(EGFR)表达水平高的胶质瘤细胞生长的导向抑制作用。方法通过结晶紫染色和3H┐亮氨酸掺入量的测定,观察基因重组TGFα┐PE40对培养脑胶质瘤U251细胞生长及其蛋白质合成的影响。结果TGFα┐PE40浓度为1~100ng/ml时,对脑胶质瘤U251细胞生长及其蛋白质的合成是呈剂量依赖性抑制。结论这表明TGFα┐PE40对脑胶质瘤U251细胞的生长具有较强的抑制作用。
Objective:The growth of glioma cells (with highly expressed epithelium growth factor receptor) suppressed by recombinant transforming growth factor αPseudomonas exotoxin fusion protein (TGFαPE40) was tested for.Methods:The effect of TGFαPE40 on cell growth and protein synthesis of U251,a glioma cell line,was observed by crystal violet staining and 3Hleucine.Results:The suppression was in a dosedependent form,when the concentration of TGFαPE40 was within 1~100 ng/ml.Conclusions:It indicates that the recombinant TGFαPE40 can obviously suppress the growth of U251 cells.
