摘要
目的:测定护肤粉刺霜中甲硝唑、氯霉素、地塞米松磷酸钠及维生素A酸的含量。方法:高效液相色谱法,以氢化可的松为内标,NovaPakC18柱,甲醇水为流动相;变换检测波长;梯度洗脱。结果:一次进样即可完成4种化学性质不同组分的含量分析。甲硝唑6.046μg·ml-1~66.51μg·ml-1,氯霉素5.872μg·ml-1~64.59μg·ml-1,地塞米松磷酸钠0.6022μg·ml-1~6.624μg·ml-1,维生素A酸0.4654μg·ml-1~5.119μg·ml-1范围内,峰面积与其浓度呈良好线性关系;平均回收率依次为96.51%,RSD=1.3%;98.74%,RSD=0.9%,96.03%,RSD=1.7%,97.61%,1.9%。结论:方法简便、快速、准确,可作为样品的检测方法。
OBJECTIVE: To separate and determine four constituents including metronidazole, chlormycetin, dexamethasone and vitamin A acid in Hufufenci Shuang. METHODS: A HPLC method was used. Chromatographic conditions included NovaPak C18 column and the mobile phase consisting of methanol and water. Hydrocortisone was used as internal standard. Alternating detection wavelength and gradient elution was used in HPLC. RESULTS: The separation and determination of four constituents having various chemical properties were accomplished by only one injecting sample. The calibration curves were linear in the range of 6.046 μg·ml-1~66.51 μg·ml-1 for metronidazole; 5.872 μg·ml-1~64.59 μg·ml-1 for chloromycetin; 0.602 2 μg·ml-1~6.624 μg·ml-1 for dexamethasone; 0.465 4 μg·ml-1~5.119 μg·ml-1 for vitamine A acid. The average recoveries of metronidazole, chloromycetin, dexamethasone and vitamin A acid were 96.51% (RSD=1.3%), 98.74% (RSD=0.9%), 96.03% (RSD=1.7%), and 97.61% (1.9%), respectively. CONCLUSION: This method was found to be simple, quick and accurate for simultaneous determination of the four constituents in Hufufenci Shuang.
