摘要
目的:优化从蛹虫草菌皮中提取、纯化虫草素方法并鉴定其纯度。方法:以虫草素提取量为指标,优化虫草素提取工艺;以虫草素的吸附与解吸特性为指标,筛选不同树脂;采用结晶法成功的获得虫草素晶体并对其纯度进行鉴定。结果:用15倍药材重量的体积分数为70%的乙醇提取3次,每次1h为最佳提取条件;D001大孔强酸离子交换树脂吸附量、吸附率最高,直接洗脱后纯度可以达到80%左右,适合虫草素的纯化;重复结晶得到虫草素晶体纯度达98%以上。结论:该方法简单,分离的虫草素纯度高,是一种可行的虫草素提取方法,有利于蛹虫草综合开发利用和虫草素大规模的工厂生产。
Objective: to optimize the technology of the extraction and purification of cordycepin from Cordyeeps militaris hulls and identify the purity of cordycepin. Methods:the yields of cordycepin in the extract were used as index to optimize the extraction process, The adsorption and desorption characteristics of cerdycepin were used to select the best resin. The pure cordycepin was obtained by recrystallization. Results: the optimum extraction technology were as follows: ratio of material to solvent 1 :15,70% alcohol to extract 3 times, lh once. By D001 macroporous strong acidic resins,the purity of cordycepin was about 80%. And by recrystallization,its purity was up to 98%. Conclusion: this study supplied a simple and convenient technology to obtain cordycepin, indicating that it was feasible to produce cordycepin from Cordyeeps militaris hulls by large-scale.
