神经节苷脂GM_1对大鼠MCAO/IR模型细胞外液氨基酸含量的影响—活体微透析研究 被引量:4

摘要 目的探讨GM1对脑缺血/再灌流的保护作用。动态观察神经节苷脂GM1对大鼠MCAO/IR模型细胞外液氨基酸类神经递质的影响。方法采用缺血1h再灌流2h模型,用活体微透析法,检测缺血皮层脑组织细胞外液内氨基酸类神经递质。结果(1)脑缺血后60minGlu上升到峰值浓度,随后很快下降,再灌流2h内未见回升;(2)GM1组各类氨基酸都明显下降。结论GM1能使包括Glu在内的多种氨基酸明显下降。 Objective To study the protective function of GM 1 in cerebral ischemia and reperfusion and effect of GM 1 on amino acid changes during rat cerebral artery occlusion and reperfusion(MCAO/IR).Methods Using 2hr ischemia/1hr reperfusion suture model,we measured extracellular amino acids collected from ischemic brain cortices by HPLC.Results 1.Glutamate increased rapidly and reached a peak level at 60 minute after ischemia;2.In GM 1 group,all determined amino acids were significantly decreased.Conclusion Our resulsts suggest that the protective function of GM 1 in brain ischemia/reperfusion may be related to the maintenance of membrane integrates of neurons.
