摘要 目的:探讨一氧化氮(Nitric Oxide,NO)供体硝普钠(sodium nitroprusside,SNP)对成纤维细胞形态及凋亡的影响。方法:将NO供体SNP制成5个不同浓度(100、200、300、400、500)μmol/L作用于成纤维细胞,观察不同时间的细胞形态变化及细胞凋亡。结果:随SNP作用浓度增高及时间延长,成纤维细胞形态呈逐渐老化状态,且数量明显减少,呈显著性负相关(y=28.943-0.858x)。成纤维细胞凋亡率300和500μmol/L组(25.67±0.36和56.38±2.18)%明显高于空白对照组(1.35±0.26)%,具有显著性差异(P<0.001)。结论:NO对成纤维细胞具有时间浓度依赖效应,可致细胞数量减少、形态老化及凋亡增加,为瘢痕防治提供了一种新的理论思路。 Objective To study the effects of sodium nitroprusside (SNP) which is the donor of Nitric Oxide (NO) on cell morphology and apoptosis of fibroblasts. Methods Prepared SNP of five different concentration(100,200,300,400,500) μmol/L for acting on the fibroblasts, then the Morphologic change and apoptosis of fibroblasts were observed dynamically. Results With the increase of concentration and elongation of acting time, the aging state of fibroblasts appeared more and more obviously, meanwhile the quantity of the cells decreased, showing significantly negative correlation (y=28.943-0.858x). The apoptosis rate of fibroblasts in the 300 and 5001Jmol/L SNP group (25.67±0.36 and 56.38±2.18 )% were obviously higher than the blank control group (1.35±0.26)%, the difference being significant (P〈 0.001). Conclusions Exogenous NO had made the number of fibroblasts decreased, the aging state seriously and the apoptosis rate increased, showing a dose and time-dependent effect. This research offers a new way of thinking on the prevention and treatment of scar .
