柱状嗜纤维菌脂多糖对鲤不同组织中离体培养淋巴细胞转化率的影响 被引量:6

摘要 从鲤胸腺、头肾和血液中分离淋巴细胞,在添加不同剂量的柱状嗜纤维菌脂多糖(LPS)的RMPI1640培养基中培养一定时间后,采用MTT比色法测定LPS对淋巴细胞的刺激指数(SI),比较了LPS对不同组织中离体培养淋巴细胞的转化率。结果表明,LPS添加量和培养时间对淋巴细胞的转化率都有明显的影响,当LPS添加量为20μg/ml,培养时间为72h,鲤各种组织中淋巴细胞的转化率达到峰值。血液中淋巴细胞转化率最高,SI达到1.77;头肾其次,SI为1.33;而胸腺中淋巴细胞的转化率最低,SI仅为1.03。 Lymphocytes were separated from thymus, head kidney and blood of carp (Cyprinus carpio) and then incubatedin RMPI1640 medium added with various dosages of Cytophaga columnaris lipopolysaccharide (LPS) for a certain period of time. Stimulation indexes of lymphocytes from the issues were determined by MTT colorimetric assay to compare thetransformation rate caused by LPS of in vitro lymphocytes.The results showed that the transformation rate of lymphocytes was remarkably affected by both the additive dosage of LPS and incubation time. The transformation rate of lymphocytes from the issues of carp reached the peak at the additive dosage of 20μg/ml LPS and 72 hours of incubation. The transformation rate of lymphocytes from blood was highest, with stimulation index (SI) of 1.77, that from head kidney was power, with SI of 1.33, and  that from thymus was lowest, with SI of only 1.03.
