高效液相色谱法测定胃癌周围淋巴结中氟尿嘧啶 被引量:4

摘要 目的:建立高效液相色谱法测定胃癌周围淋巴结中氟尿嘧啶浓度的方法。方法:以C18柱为固定相,水(pH5.8)-甲醇(95∶5)为流动相,265nm检测。结果:标准曲线方程为Y=0.01952 X-0.07016,r=0.998,线形范围18~720μg·L-1。日内及日间精密度分别为2.5%~5.7%和5.9%~7.6%。3种不同浓度的相对回收率为98.5%~103.7%,绝对回收率为70.9%~78.5%。结论:本方法简便、可靠、灵敏度高,适用于在淋巴结组织中检测氟尿嘧啶。 OBJECTIVE To develop a simple and rapid HPLC method for the determination of 5-Fu concentration in lymph node around the stomach cancer. METHODS C18 column was used the stationary phase,the water (pH 5.8) - the methyl alcohol (95 : 5) as mobile phase, UV detection wavelength was 265 nm. RESULTS The equation of standard curve was:Y = 0. 019 52X- 0. 070 16, r = 0. 998, Linear range: 18- 720μg·L^-1. Within-day precision and between-day precision were 2.5 % - 5. 7% and 5.9%- 7. 6% respectively. Relative recovery is 98. 5% - 103. 7%, and absolute recovery was 70. 9%- 78. 5% CONCLUSION This method is simple, reliable with good sensitivity.
