摘要
目的研究槲皮素(Quercetin,Que)在体外对人视网膜母细胞瘤(Retinoblastoma,RB)Y79细胞株的生长抑制和凋亡的影响。方法体外培养Y79细胞:在体外对Y79细胞经不同浓度槲皮素处理48h、72h后进行检测,用MTT法测定半数抑制浓度(IC50),观察槲皮素的细胞毒性作用,用Hoechst33342/Pl双荧光染色观察细胞核形态变化。结果槲皮素在一定浓度范围内能明显抑制Y79细胞的生长,并存在剂量-效应关系;应用MTT法计算不同浓度槲皮素作用48h、72h的半数抑制浓度(IC50)是148μmol/L、115μmol/L。Hoechst33342/Pl双荧光染色可观察到明显核固缩、凝集等,细胞凋亡的形态学改变。结论槲皮素可使视网膜母细胞瘤Y79细胞增殖抑制和凋亡诱导。
Objective To investigate the effect of Quercetin on growth of the RB cell line Y79 In vitro and the correspending mechanism involved in cell apoptosis. Methods Y79 was cultured in vitro and Inhibition rate of Quercetin on Y79 ceils was assayed after 48 and 72 hours, then studied by MTr method to measure IC50 so as to observe the cytotoxicity of Quercetin. The morphology changes of the nucleons were observed, by Hoechst 33342/P1 staining. Results Pro- liferation of Y79 cells were significantly inhibited by Quercetin in the way of dose - effect relation. After the Y79 cells were treated by different concentrations of Quercetin for 48 and 72 hours, the ICso was 148 μmol/L and 115 μmol/L by MTr experiments and distinct morphology changes of cell apoptosis such as karyopyknosis and conglomeration were observed by Hoechst 33342/P1 staining. Conclusion Quercetin inhibits proliferation of Y79 cells and induces cells specifically apeptosis.
