摘要
目的:建立柱前衍生法测定桑叶中游离氨基酸的含量。方法:以水超声提取桑叶中的氨基酸,以异硫氰酸苯酯柱前衍生,采用反相高效液相色谱法测定氨基酸的含量。结果:桑叶中含有苏氨酸、缬氨酸、脯氨酸、蛋氨酸、谷氨酸、甘氨酸、亮氨酸、苯丙氨酸、酪氨酸、天门冬氨酸、γ-氨基丁酸、丙氨酸、组氨酸、胱氨酸、色氨酸、异亮氨酸等16种游离氨基酸;其中人体必需氨基酸有7种,人体半必需氨基酸有1种,含量占氨基酸总量的20%左右。结论:本法不需要专门的氨基酸分析仪,操作简便、灵敏度高,结果准确、可靠。桑叶中氨基酸的种类齐全、含量丰富,具有很高的开发利用价值。
OBJECTIVE: To establish an HPLC method with pre- column derivation for determining the contents of free amino acids in Morus alba leaves.METHODS: The free amino acids in Morus alba leaves were ultrasonically extracted with water and determined by RP HPLC using phenyl isothiocyanate(PITC) as the pre- column derivation reagent. RESULTS: There were 16 kinds of free amino acids(threonine, amino - isovaleric acid, proline, methionine, aminoglutaminic acid, aminoacetic acid, amidocaproic acid, phenylalanine, Ganimalon, aspartic acid, γ - aminobutyric acid, alanine, histidine, dicysteine, tryptophan,isoleucine) in Morus alba leaves,of which, 7 were essential amino acids and 1 semi - dispensable amino acid, their total contents accounting for 20% of total.CONCLUSIONS: This method is simple, sensitive, accurate and reliable in results and in which no special amino acid analyzer is needed.The amino acids in Morus alba leaves are rich in variety and contents, possessing great value for development and utilization.
