摘要
目的:研究和血生络方对大鼠脑缺血再灌注损伤的保护作用,并探讨其对血管新生的作用。方法:将Wistar大鼠随机分为正常组、模型组、和血生络方组,后两组制备大鼠大脑中动脉缺血再灌注(MCAO/IR)模型,和血生络方组给予和血生络方灌胃,各组分别于1d、3d、7d、14d、21d时免疫组化法检测血管内皮生长因子(VEGF)和血管生成素-1(Ang-1)的表达。结果:模型组各时间点VEGF的表达较正常组增加(P〈0.05),在7d达到高峰,和血生络方组比模型组增加更明显,差异有显著性(P〈0.05~0.01)。模型组各时间点Ang-1的表达较正常组增加(P〈0.05),14d达到高峰,和血生络方组比模型组增加更明显,差异有显著性(P〈0.05~0.01)。结论:和血生络方可能通过增强VEGF、Ang—1的表达来促进大鼠脑缺血再灌注后的血管新生。
Obiective : To study the protective effect and the angiogenesis mechanism of Hexueshengluofang on focal cerebral ischemia/reperfusion in rats. Methods: The rats were randomly divided into normal group, model group and Hexueshengluofang group, after MCAO/IR (Middle Cerebral Artery Occulusion/ Reperfusion Injury) models were set up in latter two groups,the Hexueshengluofang group were given Hexueshengluofang, expression of VEGF and Ang-1 was detected with immunonhistochemical method after lday,3days, 7days, 14days. Results:The exression of VEGF in the Hexueshengluofang group were reached a peak on the 7th day, and significantly increased as compared with those in the model group and the normal group at each time( P 〈 0.05-0.01). The exression of Ang-1 in the Hexueshengluofang group were reached a peak on the 14th day, and significantly increased as compared with those in the model group and the normal group at each time( P 〈 0.05-0.01 ). Conclusion: Hexueshengluofang's improving ischemic stroke is carried out possibly through up regulating the expression of VEGF and Ang-1 in the cerebral tissue.
