摘要
目的观察Ang-Ⅱ对血管内皮细胞线粒体膜电位的影响及茶多酚的保护作用。方法将血管内皮细胞分为空白对照组(仅给予细胞培养液)、单纯Ang-Ⅱ组(细胞培养液中加入Ang-Ⅱ,使其终浓度为10-7mol·L-1)、Ang-Ⅱ+小剂量茶多酚组(在单纯Ang-Ⅱ组的基础上加入茶多酚,使茶多酚的终浓度为5mg·L-1)、Ang-Ⅱ+大剂量茶多酚组(在单纯Ang-Ⅱ组的基础上加入茶多酚,使茶多酚的终浓度为25mg·L-1)。用激光共聚焦显微镜测量各组细胞的线粒体膜电位水平。结果(1)Ang-Ⅱ组的线粒体膜电位显著低于空白对照组(P<0.01);(2)Ang-Ⅱ+茶多酚组的线粒体膜电位显著高于Ang-Ⅱ组(P<0.01);(3)Ang-Ⅱ+大剂量茶多酚组的线粒体膜电位水平高于Ang-Ⅱ+小剂量茶多酚组(P<0.05)。结论Ang-Ⅱ可引起血管内皮细胞线粒体膜电位的显著降低,而茶多酚可呈剂量依赖性的逆转Ang-Ⅱ的这一作用。
Aim To investigate the effect of angiotensin- Ⅱ ( Ang- Ⅱ ) on mitochondria membrane potential in vascular endothelial cells (VEC) and protection of tea polyphenols. Methods Cultured VECs were divided into 4 groups : control group ( given cell culture medium only) ; Ang- Ⅱ group ( given Ang-Ⅱ at the final concentration of 10^-7mol.L^-1) ; Ang-Ⅱ + low closage tea polyphenols group (given tea polyphenols at the final concentration of 5mg.L^-1 at the base of Ang-Ⅱ group; Ang-Ⅱ + high dosage tea polyphenols group (tea polyphenols given at the final concentration of 25mg.L^-1 at the base of Ang-Ⅱ group). Mitochondria membrane potential was measured by laser scanning confocal microscopy. Results ( 1 ) Mitochondria membrane potential of Ang- Ⅱ group was significantly lower than that of control group ( P 〈 0.01 ) ; (2) mitochondria membrane potential of Ang- Ⅱ + tea polyphenols group was significantly higher than that of Ang- Ⅱ group ( P 〈 0.01 ) ; ( 3 ) mitochondria membrane potential of Ang- Ⅱ + tea polyphenols group was higher than that of Ang- Ⅱ group ( P 〈 0.05 ). Conclusion Ang- Ⅱ induce obvious changes of mitochondria membrane potential in VECs and tea polyphenols inhibits it in a dose-dependent manner.
