摘要
用还原型烟酰胺腺嘌呤二核苷酸黄递酶(NADPH-diaphorase)组织化学方法,检测空间辩别性学习记忆模型大鼠和对照大鼠的海马结构内一氧化氮合酶(NOS)的活性,阳性细胞的分布、形态和数量等形态学指标.结果:(1)模型大鼠海马结构内NOS阳性神经元的酶反应产物比对照大鼠的明显减少;(2)模型大鼠和对照大鼠海马结构内NOS阳性神经元的分布未见组间差异,但均显示出明显的区间差异,即从CA4区向CA1区逐渐增多;(3)模型大鼠和对照大鼠海马结构内的NOS阳性神经元的数量显示出明显的组间差异,即模型组比对照组的明显减少;(4)模型大鼠和对照大鼠海马结构内的NOS阳性神经元形态主要有锥体形、颗粒形和纺锤形三种,但未见组间差异.据此,可认为大鼠海马结构内NOS阳性神经元在空间辨别性学习记忆活动时发生了形态学可塑性.
The method of NADPH histochemical method was used to show the neurons containing NOS in the hippocampal formation and then conut optical density (OD) of reactive product with image pattern analysis instrument and observe the distribution,morphology,quantity of neurons containing NOS under light microscopy.The results show 1) reactive product’s OD titre of the model animal less than that of the control animal and the difference is statistically significant (P<0.01).2) The distribution of the neuron containing NOS in the hippocampal formation show a obvious regional difference in the model animals and contral animals.In the hippocampus,the most neurons in number are in CA1 region and the least in the CA4 region,the median in the CA3 and CA2 regions.In the dentate gyrus,monolayer neuron is in lateral and medical crest.The stratified neurons are in the conjugation of the medial and lateral crests.In the subculium,the neurons are more than that inthe hippocampus and dentate gyrus.there is no difference in distrbution of the neurons containing NOS between goups.3) The morphologies of the neuron containing NOS in the hippocampal formation in the model animals and contral animals show three kinds of morphology the first is pyramidal or triangle that has longer dendrite and more branch and distribute in the mokecular and lacuma layer of the hippocampus;the second is round or oval that has shorter dendrite without branch,the distributes in dentateg yrus,the third is fusiform with the branch parallelnig brain surface and distributes in the orients layer.In the control animals,the distribution of the neuroms containing NOS is the same as that of the model animals.4) The number of the neurou containing NOS in the model animals is less than control animals.As mentioned above,the author believes that the morphological plasticity happens on the neurous containing NOS in the rat hippocamal formations induced by the activity of learning and memory,it suggests NO may relate to learning and memory.
