摘要 目的探讨犬骨髓间充质干细胞(BMMSCs)向血管内皮样细胞分化的机制。方法骨髓穿刺获取犬骨髓,提取单个核细胞,取第2~4代细胞用含VEGF的培养液诱导,对细胞进行形态学观察和相关抗原的免疫荧光鉴定。结果BMMSCs在含VEGF的培养液中诱导,2周后外观呈现鹅卵石样,免疫表型显示vWF阳性表达,透射电镜可见细胞具有血管内皮细胞的特点。结论骨髓间充质干细胞可在体外分化为具有血管内皮细胞特性的细胞。 Objective To investigate the induction of BMMSCs into vascular endothelium-like cells (VELCs) in vitro. Methods The canine bone marrow was aspirated from sternum. BMMSCs were isolated and purified by density gradient centrifugation and adherion to the culture plastic. VELCs derived from BMMSCs were cultured in DMEM-LG with VEGF and observed by morphology and immunofluorescence. Results VELCs were cultured and sub-cultured successfully. VELCs showed cobble stone-like morphology and sufficient cell organs. Immunofluorescence results showed vWF of VELCs was positive. Conclusions BMMSCs can be induced into VELCs with the characteristic of vascular endothelial cells.
