摘要
利用PCR方法,从拟南芥中克隆了865 bp的逆境诱导型启动子Prd29A。利用农杆菌介导的瞬时表达方法,结合GUS染色,在豆科植物百脉根的幼苗中进行活性鉴定。结果表明:启动子Prd29A在正常情况下不表现活性,而在逆境(高渗、高盐)和ABA诱导下活性很高,这为该启动子在百脉根等豆科植物中的应用提供了依据。
Prd29A, A 865 bp in length of stress induced promoter, was cloned by PCR from A. thaliana. The activity of the promoter was evaluated in the leguminous plant Lotus cirniculatus L. by transient expression via agrobacterium-mediated method united with GUS assay in young seedlings. The results indicated that the promoter Prd29A performed no activity under normal condition, but high activity induced by stresses (high salinity, osmotic) and ABA. These results could facilitate the improvement of leguminous plants transformation with Prd29A.
