干扰素-α对CCL13/HBx细胞生长及侵袭转移潜能的抑制作用

Inhibitory effects of interferon-α on growth and invasion potentials of hepatocellular carcinoma induced by hepatitis B virus X protein

摘要 目的观察干扰素-α对恶性转化的CCL13/HBx细胞的生长和侵袭转移潜能的抑制效应。方法采脂质体法将pcDNA3.1-HBx质粒转入CCL13细胞,将CCL13/HBx细胞分别在普通条件下培养或与干扰素-α共培养,采用绘制生长曲线、平板集落形成实验、划痕愈合实验、体外侵袭力、体内裸鼠成瘤实验分别检测CCL13/HBx、INF—α-CCL13/HBx、CCL13/pcDNA3.1细胞在体内、体外的生长及运动迁移能力的差异。结果pcDNA3.1-HBx质粒转染CCL13细胞后在该细胞中稳定表达,CCL13/HBx细胞生长明显快于INF—α—CCL13/HBx和CCL13/pcDNA3.1细胞,克隆生成率明显增高(31.2%比10.8%比12.4%,P〈0.05),划痕愈合能力明显增强,穿过人工基底膜的细胞数明显增多[(41.6±3.1)/HP比(7.4±1.2)/HP比(9.2±1.6)/HP,P〈0.05]。干扰素-α治疗组CCL13/HBx细胞裸鼠皮下成瘤体积减小[(2.4±0.2)cm^3比(4.2±0.3)cm^3,P〈0.05]。结论CCL13/HBx细胞较CCL13/pcDNA3.1细胞具有更强的生长和侵袭转移潜能,干扰素-α能明显抑制HBx蛋白所诱导的细胞恶性表型。 Objective To explore the inhibitory effects of interferon-alpha (INF-α) on growth and invasion potentiald of hepatoceilular carcinoma induced by hepatitis B virus X protein (HBx). Methods The pcDNA3. 1-HBx plasmid was transfected into CCL13 ceils by Lipofectamine in vitro, and CCL13/HBx was co-cultured with INF-α. Cell survival growth curve and elonogenicity assay were used to test the growth potential of CCL13/pcDNA3. 1, CCL13/HBx and IFN-α-CCL13/HBx in vitro. Tumor growth assay in nude mice was used to detect the growth potentials of them in vivo, and wound-healing and transwell migration assays were used to detect their invasive ability. Results The human liver cell lines CCL13 containing stabilizing HBx gene expression had the characterizations of malignant ceils, they grew faster than INF-α-CCL13/HBx and CCL13/pcDNA3.1 cell lines obviously, had stronger colony- forming efficiency ( 31.2% vs 10.8% vs 12.4%, P 〈 0.05 ) and wound healing capability. Their number of ceils which penetrated the lower surface of the matrigel-coater filters was more than the other two cell lines [ (41.6 ± 3.1 )/HP vs (7.4 ±1.2)/HP vs (9.2 ± 1.6)/HP, P 〈 0.05 ]. But their tumorigenity in nude was lower when treated with INF-α [ ( 2.4 ± 0.2) cm^3 vs (4.2± 0.3 ) cm^3, P 〈 0.05 ]. Conclusion INF-α can restrain the growth and invasive potentials induced by HBx protein in hepatocellular carcinoma cells,which provided an experimental basis for hepatocellular carcinoma therapy.
