五羟黄酮对裸鼠人肝癌细胞Survivin基因表达的抑制作用 被引量:1

The inhibitory effect of quercetin on trisphosphate inositol and Survivin gene expression in hepatocellular carcinoma of nude mice

摘要 目的观察三磷酸肌醇(IP3)和Survivin基因表达变化在五羟黄酮(quercetin)治疗裸鼠移植入肝癌中的作用。方法以裸鼠移植入肝癌为对照,对照组腹腔注入含0.04%DMSO的RPMI 1640培养基0.05ml/g,Quercetin治疗组腹腔注入quercetin每日50mg/kg体重,3周后观察肝癌增长情况,3周后观察肝癌增长情况,并应用同位素试剂盒检测肝癌组织IP3含量,RT—PCR分析癌组织Survivin mRNA表达,Western blot分析肝癌组织Survivin蛋白表达。结果治疗组肝癌体积和重量均显著低于对照组[体积(15.8±10.1)mm^3比(52.3±26.5)mm^3,重量(44.8±10.4)mg比(91.3±31.4)mg],IP3含量显著低于对照组[(13.4±1.4)pmol/mg protein比(35.3±6.6)pmol/mg protein],Survivin mRNA表达低于对照组但差异无统计学意义[RI(灰度与面积之积的相对强度)0.60±0.06比0.79±0.19](P〉0.05),Survivin蛋白表达显著低于对照组[RI(灰度与面积之积的相对强度)1.23±0.25比6.69±1.80]。结论Quercetin能减少IP3生成,下调肝癌组织Survivin基因表达,抑制裸鼠移植入肝癌增长。 Objective To investigate the implication of 1,4,5 - trisphosphate inositol (IP3) and Survivin gene expression in the treatment of hepatocellular carcinoma of nude mice by quercetin. Methods Animals with hepatocellular carcinoma were treated with quercetin 1 mg/(kgoday) (i. p) for 3 weeks. The volume and weight of tumaors were measured, and IP3, Survivin mRNA and Survivin protein were assayed by IP3 -[ ^3H ] Birtrak assay, RT-PCR and Western blotting, respectively. Results The tumor volume and weight of animals treated with quercetin were less than controls [ Volume (5.8 ± 10.1 ) mm^3 vs (52.3 ±26.5) mm^3 ; weight (44.8 ± 10.4) mg vs (91.3 ± 31.4) mg], IP3 continent was lower than that of controls [ ( 13.4 ± 1.4) pmol/mg protein vs ( 35.3 ± 6.6 ) pmol/mg protein ] , and the Survivin mRNA expression was lower in the group treated with quercetin than that of controls but had no significant difference [ RI 0.60 ± 0.06 vs 0.79 ± 0. 19 ] ( P 〉 0.05 ). Survivin mRNA expression was lower in the group treated with quercetin than that of controls ( 1.23 ± 0.25 vs 6.69 ± 1.80). Conclusion Quercetin can inhibit the growth of hepatocellular carcinoma tansplanted into liver of nude mice by reducing IP3 production and down-regulating Survivin gene expression.
