摘要
目的探讨大鼠肾缺血不同时段及预适应(IP)再灌注损伤(IR)I中肝细胞生长因子(HGF)的表达意义.方法将热缺血大鼠随机分成4组:分别钳夹左肾蒂0、30、60、90min,再灌注24h后取标本,IP分为两组,在热缺血之前先做预处理然后分别与IRI60min组及IRI90min组相对比.各组均检测血肌酐和尿素氮,左肾组织HE染色行肾小管评分,ELISA法检测血清的HGF水平值.结果缺血60min组及90min组对肾功能损伤都严重.IP1组与IRI60min组的各项指标有统计学意义.HGF在60min组和IP1组与对照组在统计学上有统计学意义.结论IRI60min组是IRI研究的最佳模型,IP1组能有效减轻缺血再灌注所致的肾脏的损伤,HGF参与IRI肾功能的恢复,比肾功能指标更敏感.
Objective To investigate the significance effective of HGF in the rat renal different ischemic times and ischemic preconditioning reperfusion injury. Methods Ischemia rats were divided into four groups: clamp left kidney were clamped 0 min, 30 rain, 60 min, 90 min, according to reperfusion after 24 h, we detected ischemia group renal function of the degree of serious injury. IP were divided into two groups, ischemic preconditioning before heat ischemia were compared with 60 min and 90 min group. Each group left kidney organizations werestained HE and pathomorphology analysised, The serum of HGF level the rats in each group was tested by ELISA. Results 60 min and 90 min groups of renal injury were serious. IP1 was compared with 60 min group and showed significant difference; HGF in 60 min and IPlwere compared with the control group and showed statistical significance. Conclusion The 60 min group is the best model of IRI. IP1 can effectively reduce IRI. HGF participates in the restoration of renal IRI and is more sensitive than the indicator of renal function.
