肺泡Ⅱ型上皮细胞的原代培养及生物学特性比较研究 被引量:5

摘要 目的:建立大鼠肺泡Ⅱ型上皮细胞(AECⅡ)的改良体外原代培养法,并对其生物学特性进行比较研究。方法:AECⅡ用胰蛋白酶和胶原酶消化法分离后,接种于大鼠免疫球蛋白G(IgG)包被的培养瓶黏附纯化。将获得的AECⅡ用碱性磷酸酶(AKP)、电镜和肺表面活性物质相关蛋白A(SPA)免疫细胞化学方法进行鉴定,并对它的SPA表达和转染特性与AECⅡ来源A549细胞进行比较研究。结果:胰蛋白酶和胶原酶消化加IgG黏附纯化后所得的AECⅡ,产量为2×107-3×107,纯度为75%-84%,细胞活力93%以上,AKP染色快捷简便。AECⅡ同A549细胞相比具有某些不同的特性。结论:采用IgG黏附纯化能提高AECⅡ纯度,AKP染色简便实用。对于AECⅡ细胞的某些特性研究发现其并不能完全用A549细胞系来代替。
