摘要
目的从BALB/C小鼠的早期胚胎中分离和培养胚胎干细胞(ES细胞),并对其生物学特性进行初步鉴定。方法收集BALB/C小鼠3.5d胚龄的囊胚,分离内细胞团(ICM)细胞进行培养,以小鼠原代胚胎成纤维细胞(PMEF)作为饲养层,细胞扩增传代,观察集落的生长情况并通过碱性磷酸酶(AKP)染色对细胞集落进行初步鉴定。结果ES细胞呈集落样生长,AKP染色阳性,符合小鼠ES细胞的一些特性。结论BALB/C小鼠囊胚在原代小鼠胚胎成纤维细胞饲养层上可以发育成ES细胞。
Objective To isolate and culture embryonic stem(ES) cells from embryos of BALB/C mice, and preliminarily identify their bionomics. Method Collected the blastocysts of 3.5d from BALB/C mice, inner cell mass (ICM)were isolated and cultured on the primary mouse embryonic fibroblast (PMEF)cell feeder layers. The ES cells were preliminarily identified by characterized morphology, alkaline phosphatase activity assay. Results The ES cells were grown on the PMEF feeder layers and possess colony morphology, alkaline phosphatase positive reaction, ES cells were satisfied some traits of murine ES cell line. Conclusions The blastocysts from BALB/C mice can be cultured on the PMEF feeder layers and developed into ES cells.
