摘要
文献报道的去三苯甲基的方法,除个别外,大都易影响其它酸敏感基团.本文报道一个反应条件温和,选择性好的方法,不仅在异亚丙基、叔丁基二甲硅基、甲氧甲基(MOM)、甲氧乙氧甲基(MEM)、苷键上的苯甲酰基等酸敏感基团存在时有选择性,而且当五碳呋喃糖5-OH与1-位半缩醛羟基同时具有三苯甲基时,对脱去后者,也有一定的选择性.
A method of mild and selective detritylation is reported.The sugar derivatives 1a,2a, 3a, 4a, 5a, with a variety of protectine groups, were detritylated with p-TsOH(0.1—0.2 equiv.)in CH_2Cl_2 at room temperature.Acid sensitive groups such as isopropylidene,methoxyl methyl (MOM),methoxyl ethoxyl methyl (MEM),t-butyl dimethyl silyl (TBDMS),glycosidic benzoyl survived. With pentose derivatives 6a, 7a containing two trityls at 1-,5-position,1-trityloxy can be cleaved selectively at lower reacton temperature and shorter reaction time.Yields range from 53 to 90%.
