日本血吸虫成虫生殖系统的激光共聚焦显微镜形态学观察 被引量:2

Morphological Observation on the Reproductive System of Adult Schistosoma japonicum under Confocal Laser Scanning Microscopy

摘要 BALB/c小鼠经皮感染日本血吸虫尾蚴(40±2条/只)35d后剖杀,获取成虫,经固定、染色、脱色、脱水、透明和封片等处理制成整装玻片标本,用激光共聚焦显微镜观察。显微镜下日本血吸虫的整体形态以及雄虫生殖系统中的睾丸、贮精囊和生殖孔,雌虫生殖系统中的卵巢、卵黄腺、输卵管、卵黄管、受精囊、卵模、梅氏腺、子宫、雌性生殖孔以及虫卵等形态结构清晰。激光共聚焦显微镜技术可较好地观察日本血吸虫虫体各器官组织和细胞结构。 BABL/c mice were infected with Schistosomajaponicum cercariae (40±2 per mouse) through abdominal skin. Mice were sacrificed after 35 days to acquire the adult worms which were then fixed, stained, clarified, dehydrated and mounted. The specimens were observed under the eonfocal laser scanning microscope. The overall morphology of the adult worms was displayed distinctly, especially the testieular lobes, seminal vesicle and genital pore of the male, reproductive system, and the ovary, vitelline glands, oviduct, vitelline duct, seminal receptacle, ootype, mehlis gland, uterus, genital pore and eggs of female reproductive system. The confocal laser scanning microscopy is an alternative method to research organs, tissues and cell structure of sehistosome worm.
