丝裂素活化蛋白激酶在冠心病患者冬眠心肌中的表达

Expressions of MAPKs in Hibernating Myocardium of Patients with Coronary Artery Disease

摘要 目的:探讨冬眠心肌细胞内丝裂素活化蛋白激酶(MAPKs)水平的变化和意义。方法:选择10例行冠脉旁路移植术(coronary artery bypass graft,CABG)的冠心病患者,术前1周内用多巴酚丁胺超声负荷试验(dobutamine stress echocardiography,DSE)结合多普勒组织成像(doppler tissue image,DTI)确定冬眠心肌及正常心肌的存在部位,CABG术中根据检测结果进行取材(分别取正常心肌和冬眠心肌),并经电镜证实。取材心肌用免疫印迹法检测磷酸化的ERK、JNK和P38的表达情况,分析冬眠心肌与正常心肌磷酸化ERK、JNK和P38是否存在差异。结果:冬眠心肌细胞内磷酸化ERK、JNK和P38水平均较正常心肌高,依次为7811.6±1094.4比6421.0±766.0、9351.6±645.5比6179.0±712.6和10164.9±536.3比9589.5±586.3,P<0.05。结论:冬眠心肌中MAPKs水平升高,可能参与冬眠心肌的形成。 Objective: To observe expressions and significance of MAPKs (mitogen activated protein kinases)in hibernating myocardium (HM). Methods:Ten patients with coronary artery disease (CAD) scheduled for bypass surgery underwent preoperative dobutamine stress echocardiography (DSE) and doppler tissue image (DTI) to check the position of HM. The normal myocardium(NM) and the HM were obtained in operation,confirmed by electron microscope. Then P-ERK,P-JNK and P-P38 were detected by western blot. Results: Expressions of P-ERK,P-JNK and P-P38 were significantly increased in HM compared with normalmyocardium 7 811.6± 1 094.4 vs. 6 421.0±66.0, 9 351.6±45.5 vs. 6 179.0±712.6 and 10 164.9±536.3 vs. 9 589.5 ± 586.3, respectively all P〈0.05. Conclusion: It increased for expressions of MAPKs in HM, and probably take part in the process of HM formation.
