摘要
目的研究β-防御素-1mRNA在正常大鼠体内的存在情况和基因顺序情况,为临床调节和应用β-防御素-1提供重要的信息。方法从大鼠不同器官和组织的匀浆中提取总RNA,合成第1链cDNA,利用PCR扩增大鼠β-防御素-1的DNA片段,PCR产物经凝胶电泳后观察并测序。结果遍及全身的21种器官和组织中,气管、心脏、肾脏、子宫、卵巢、睾丸、肝脏、胃、小肠、胸腺、脑和皮肤等12种器官或组织中能检测到β-防御素-1的mRNA,其中,在气管、心脏、肾脏、胃、卵巢和皮肤中较为丰富。DNA测序确定PCR产物为防御素,且与已知的防御素结构完全一致。结论β-防御素-1的mRNA在正常大鼠体内多个器官和组织中都存在,提示它在机体内控制细菌感染的作用较为广泛,它的结构高度保留。
Objective To investigate the existence of β-Defensins-1 mRNA and its cDNA sequence in normal rats,for providing important information about β-Defensins-1 in clinical application.Methods The total RNA were extracted from organs and tissues homogenates. Then the first-strand cDNA was synthesized and β-Defensins-1 DNA was amplified by PCR. The PCR product was tested by gel electrophoresis and sequenced. Results β-Defensins-1 mRNA were found in trachea , heart , kidney , uterus , ovary , testis , liver , stomach , small intestine,thymus, brain and skin of 21 various organs and tissues. Among them,β-Defensins-1 mRNA content was relatively rich in trachea,heart,kidney,stomach, ovary and skin. DNA sequence proved that the PCR product was β-Defensins-1, and its structure were identic. Conclusion β-Defensins-1, mRNA present in many organs and tissues of rats,which indicates that the role of β-Defensins-1 in host defense of bacteria infection is relatively extensive and its structure is highly conserved.
