2型糖尿病ecNOS基因27bpVNTR多态性与颈动脉内膜中层厚度的关系

摘要 目的研究2型糖尿病(T2DM)内皮性一氧化氮合酶(ecNOS)基因27bp数目可变的串联重复序列(VNTR)多态性与颈动脉内膜中层厚度(IMT)的关系。方法选择昆明地区汉族无血缘关系的143例T2DM患者和80例健康对照(NC)者,采用PCR技术检测ecNOS基因27bpVNTR多态性,检测相关临床指标,采用高频超声多普勒测定IMT。结果T2DM患者含有4a(4次重复系列)等位基因的频率为0.084,T2DM组与NC组的颈动脉IMT有显著差异,ecNOS4a+与ecNOS4a-组间血浆TG水平的差异有统计学意义,而两组间IMT相比差异无统计学意义。结论ecNOS4a等位基因与T2DM患者的高TG水平有关,与颈动脉IMT可能无关。 Objective To study the distribution of variable number of tandem repeats (VNTR) polymorphism of ecNOS (endothelial constitutive nitric oxide synthase) gene in Kunming Han Chinese subjects with type 2 diabetes mellitus and to investigate the correlation of the polymorphism of ecNOS with intima-media thickness(IMT). Methods Clinical indicators of unrelated Kunming Han Chinese subjects with type 2 diabetes (n = 143) and healthy control(n= 80) were investigated, and the genotypes of ecNOS were assessed by PCR. The IMT, which can be used to estimate early stages of artherosclerosis, was measured by high-resolution ultrasonography. Results Allele frequency of the 4a was 0. 084 in Kunming Han Chinese subjects. Serum triglyceride level was significantly higher in subjects with 4a allele, and the IMT values did not show any significant difference among patients with various ecNOS genotypes. Conclusions The eeNOS 4a allele is correlated with serum triglyceride level but not with IMT value.
