摘要
目的:提高对骨肉瘤免疫表型特别是浸润转移相关表型的认识及揭示骨肉瘤高转移率的部分有关可能机理,为临床提供有可能成为评价骨肉瘤患者预后的新的参考指标。方法:采用免疫细胞化学方法检测OS-732细胞系10种与转移相关的抗原表达。结果:该细胞系除p53-Pab1801染色为阴性外,其它9项均为阳性。结论:两种基质降解酶-Cath-D和MMP-2的阳性表达,推测它们对骨肉瘤的浸润转移有一定的促进作用,并可能成为骨肉瘤的两个转移表型:p53-D01和p53-Pab240蛋白的阳性表达间接说明p53基因突变的可能,不仅进一步验证骨肉瘤中p53突变的发生率高,而且综合PCNA的强阳性、BMP和上述两种酶的阳性表达,推测本细胞系具有较高的浸润能力和转移潜能,为进一步探讨骨肉瘤的高转移机理提供了一定的实验基础。
Objective: To investigate the characteristics of the invasion and metastasisrelated immunophenotype of human osteosarcoma cells and possible mechanism of its metastasis and provide the possible indices for evaluating the prognosis of patients with osteosarcoma in clinical practice. Methods: The expression of 10 metastasisrelated antigens in a human osteosarcoma cell line OS732 was determined with immunocytochemistry. Results: In the cell line, only the antigen p53Pab1801 was negatively stained. Conclusion: The cell line is of high infiltrative capacity and metastatic potential. This provides some experimental evidences for further exploration of the reason for the high metastatic rate of osteosarcoma.
