大肠杆菌LTb蛋白单克隆抗体的制备和鉴定

摘要 将大肠杆菌不耐热肠毒素LTb基因去除信号肽后与载体pPIC9K连接,电转化入酵母菌SMD1168中进行诱导表达。用纯化的LTb表达蛋白免疫8周龄BALB/c鼠3次后,取脾细胞与SP2/0骨髓瘤细胞进行融合后,以间接ELISA和有限稀释法进行筛选,获得4株能稳定分泌抗大肠杆菌LTb单克隆抗体的杂交瘤细胞株,该株单克隆抗体命名为1F2,2C2,3D6,4H6株。经Western blotting分析,这4株均能与大肠杆菌LTb表达蛋白产生特异性反应。抗体亚型鉴定表明,1F2,2C2,3D6,4H6株均为IgG2a型。成功获得单克隆抗体为今后LTb蛋白的深入研究以及抗原表位分析提供有益价值。
