摘要 应用D-GalN+ET复制急性肝衰竭(AHF)动物模型,检测肝衰竭鼠血清中肿瘤坏死因子(TNF)含量及肝组织匀浆中磷脂酶A2(PLA2))活性,探讨二者在AHF时的作用。结果发现,AHF组鼠血清中TNF含量及肝组织匀浆中PLA2活性明显高于正常对照组(P<0.05,P<0.01),提示TNF可能激活PLA2,后者与TNF所致的肝损伤有关。 To investigate the roles of tumor necrosis factor(TNF)and phospholipase A 2(PLA 2)in the liver injury and their relation,D-Galactosamine(D-Ga1N)1 100mg/kg plus endotoxin(ET)20μg/each rat were administered to induce acute hepatic failure(AHF)in rat,and the serum TNF level,PLA 2 activity in the live homogenate were measured.The results showed that the serum TNF level,PLA 2 activity in the liver homogenate were significantly higher in AHF group than that of normal group(P<0.05,P<0.01).Our results suggested that TNF and PLA 2 were involved in the pathogenesis of AHF induced by D-Ga1N+ET.It is possible that TNF mediated liver injury by activiting PLA 2.
