摘要
本文用3H-TdR掺入试验发现空肠弯曲菌43KD热休克蛋白(HSP43)能显著促使小鼠脾淋巴细胞活化,并呈剂量依赖关系。这种活化作用不但能被特异性针对HSP43的二株单抗明显抑制,而且也能被针对HSP60家族保守区序列的二株单抗所抑制。去除巨噬细胞同样也能抑制HSP43活化作用。用单抗和补体的特异性杀伤所致T细胞亚群缺失试验发现HSP43主要活化CD4+T细胞。本文结果提示空肠弯曲菌HSP43可能具有某些超抗原特性,这对解释HSP诱导自身免疫病具有一定意义。
In the experiment of murine splenic lymphocytes transformation,43KD heat shock protein(HSP43)of campylobacter jejuni,strain CJ-S131,was found to be capable of polyclonal activation.Uptake of3H-TdR transformation was linearly correlated with dosage of HSP43.Monoclonal antibodies IIH9,ML-30,CJ-1 and CJ-2 specific for widely separated linear amino acid epitopes of HSP43 strongly inhibited the activation of HSP43 on T cells,percentage of inhibition from 30% to 79% when 1:100 dilution of antibodies added into the culture.The activation of T cells by HSP43 was also strongly inhibited by removal of macrophages from lymphocytes population.Depletion of lymphocyte subset with monoclonal antibodies against CD3,CD4,CD8 and complement showed that lymphocyte subset activated by HSP43 was CD4+T cells.These results suggested that HSP43 of campylobacter jejuni might have some features of a superantigen,and also gave us an insight into the role of heat shock proteins in autoimmune diseases.
