摘要
利用启动子探针型载体pSUPV2首次从诸葛菜总DNA中克隆了7个具有启动功能的DNA片段。转化E.coli表明最高卡那霉素(kna)抗性为140μg/ml,最低为20μg/ml。含启动子片段的7个重组质粒分别命名为pSUPZ1-7。Southren印迹表明pSUPZ6对Kna的抗性功能来源于诸葛菜的10kb、5kb和3.9kb片段。
For the first time,seven DNA fragments having the ability of promotion were cloned from total DNA of O.violacecus with promoter probe vecter pSUPV2. The result of transforming E.coli showed that the highest kanamycin resistance level was 140μg/ml,and the lowest 20μg/ml.The seven recombinant plasmids with promoter fragments were named pSU P21-7.According to the result of Southern blot,the resistivity of pSUPZ6 was from the three DNA fragments of O.violaceccs with the MW.of 10kb,5kb and 3 9kb,respectively.
