摘要
目的:建立HPLC法测定神农香菊中绿原酸、木犀草素的含量的方法。方法:采用高效液相色谱法,DIONEX-AC-CLAIM.120C18(250mm×4.6mm,5μm)色谱柱;以磷酸盐缓冲溶液(pH2.5)-甲醇(梯度洗脱)为流动相;流速为0.5mL·min^-1;柱温为35℃;检测波长为350nm。结果:绿原酸在0.024-0.192μg范围内呈线形关系,r=0.9994,平均回收率为98.3%,RSD为1.2%;木犀草素在0.058~0.464μg范围内呈线形关系,r=0.9998,平均回收率为97.3%,RSD为1.8%。结论:本方法简便,重复性好,可用于神农香菊原药材质量标准。
OBJECTIVE To establish a method for the determination of chlorogenic acid and luteolin in dendranthema indicum with HPLC. METHODS DIONEX-ACCLAIM. 120 C18 column(250 mm × 4. 6 mm, 5 μm)was used, the mobile phase consisted of phosphate buffer solution (pH 2. 5) and methanol with gradient elution; flow rate was 0. 5 mL·min^-1 ;column temperature was 35℃ ; the detective wavelength was 350nm. RESULTS The linear range of chlorogenic acid was 0. 024 μg to 0. 192μg,r= 0. 999 4,the average recovery was 98. 3% with RSD of 1.2%. The range of luteolin was 0. 058 μg to 0. 464μg, r = 0. 999 8, the average recovery was 97. 3 %, RSD was 1.8 %. CONCLUSION The method is stable, reliable, precise and can be used for the quality standard of Dendranthema indicum.
