摘要
采用免疫组化ABC法观察各型大肠癌、大肠绒毛状乳头状瘤和癌切端大肠粘膜组织蛋白酶D的分布、超微结构变化及其与Ⅰ、Ⅱ型胶原关系。结果提示:第Ⅲ型粘液细胞癌(印戒细胞场)组织蛋白酶D分布于胞浆,所形成的粘液池内粘液亦呈阳性,且靠近印戒细胞周围更强。管状腺癌胞核丰富者多为阴性,低分化腺场胞浆丰富者则组织蛋白酶D弥漫于胞浆。粘液池内胶原溶解、消失。电镜下,可见第Ⅲ型粘液细胞场分泌功能旺盛,癌细胞附近亦有间质胶原溶解。提示组织蛋白酶D在大肠癌浸润中能降解间质Ⅰ、Ⅲ型胶原,故粘液细胞癌浸润性强与该酶活性有关。
The distrbution of Cathopsin D and type I collagen fibera in various typea of adenocarcinoma, villous papillary adenoma and normal mucosa of human coiorectum were investigated by means of immunohistochemical and electronmicroscopical techniques. The results showed that the Cathepsin D distributed in the cytoplasm of type Ⅲmucinous cell carcinoma as well as in the mucous pools of extracellular stroma. The cathepsin D also presented in the cytoplasm of poorly - differentiated adenocarcinoma with rich cytoplasm, whereas the cathepsin D usually was negativc in the tubular adenocarcinoma having more nuclei. Where the mucoua infiltrated, the collagen fibers, smooth muscles and vessels were lysed. The electron microscopc results showed that the secretion function of the type Ⅲmucinous cell carcinoma was high and the collagen fibers adjacent to the type Ⅲ mucinous cell carcinoma was lysed.So the present study suggested that the cathepsin D is able to degrade type mucinous cell carcinoma was lysed.So the present study suggested that the cathepsin D is able to degrade type I and Ⅲ collagen fibers in extracellular stroma and faciliates cancer invasion.
