摘要
目的:检查转化生长因子β1(TGFβ1)、ACTH对碱性成纤维细胞生长因子(bFGF)受体的影响作用。方法:肾上腺皮质束、网状带细胞(BAC细胞)的基础培养;125IbFGF受体特异性结合标记;Scatchard分析。结果:2ng/mlTGFβ1处理细胞24h,每个细胞上高亲和力受体6500个(KD=3.1511×10-11mol/L),低亲和力受体数140000个(KD=1.6277×10-9mol/L)。ACTH处理组细胞的高、低亲和力受体分别是7600和95000个/细胞(KD分别为3.6098×10-11mol/L和1.4012×10-9mol/L)。结论:TGFβ1、ACTH均可增加BAC细胞的bFGF受体数目;但二者在刺激强度和选择性上不同。TGFβ1、ACTH这对激素、因子对bFGF受体的不同调节作用可能与它们BAC细胞不同的生物学作用有关。
Objective:To examine the influence of transforming growth factor β 1(TGFβ 1) and ACTH on basic fibroblast growth factor(bFGF) receptors of bovine adrenocortical fascilata reticularis cells (BAC cells).Methods:BAC cell culture in primary condition; 125 I bFGF receptor specific binding labeling;Scatchard analysis.Results:High and low affinity receptors were identified,for the cells that were treated with TGFβ 1(2 ng/ml),high and low affinity bFGF receptors were 6 500 sites/cell(K D=3.1511×10 -11 mol/L) and 140 000 sites/cell(K D=1.627 7×10 -9 mol/L) respeectively.For the cells that were treated with ACTH(3.0×10 -7 mol),high and low affinity bFGF receptors were 7 600 and 95 000 sites per cell respectively(K D=3.609 8×10 -11 mol/L and K D=1.401 2×10 -9 mol/L correspondingly).Conclusions:The number of bFGF receptors was increased under the influence of TGFβ 1 and ACTH,a detectable difference in strenth and selectivity was observed.The difference in effects on BAC cells may be due to their difference in biological activities between TGFβ 1 and ACTH.
