用吸收光谱法检测三种烷化剂与DNA的结合 被引量:6

DETECTION OF BINDING OF THREE ALKYLANTS WITH DNA USING ABSORPTION UV SPECTROPHOTOMETRY

摘要 DNA加合物形成可作为DAN损伤的指标。应用DNA吸收光谱移位法,对3种化学物与DNA的结合作了检测。结果表明叠氮钠和甲磺酸甲酯可与DNA结合,而二甲基亚硝胺未能显示结合。结论指出,本研究中所用的测试系统可能只适用于检测直接诱变物,而不适用于间接诱变物。尽管如此,它将还是一种快速筛检诱变物的有用方法。 Formation of DNA adduct may serve as an indicator of DNA damage.Detection of binding of 3 chemicals with DNA was conducted based on a shift of DNA absorption spectrum.The results showed that sodium azide and methyl methanesulfonate were bound with DNA,but not dimethylnitrosamine.It was concluded that the system used in this study may be applied for detecting direct mutagens,but not indirect mutagens,and will be useful method for quick screening of mutagens.
