摘要
为探讨重组合异种骨(RBX)移植对机体免疫功能的影响,以RBX、牛BMP、经处理的同源松质骨粒、羟基磷灰石移植建立4组BALB/C小鼠股后肌袋模型.术后2、7、14、28、42天杀鼠取脾细胞.留取上清液.3H-TdR掺入法时体内植骨及体外同源抗原刺激后IL-2生物学活性进行了研究.结果表明:①RBX(BMP)移植后2天IL-2一过性升高;②术后7、14天IL-2水平显著降低;③体外抗原刺激各时间点3H-TdR掺入cpm值明显降低,且与抗原量呈剂量依赖关系.上述结果提示,RBX移植对T淋巴细胞和IL-2诱导CTLL增殖有明显的抑制效应,可能对IL-2也有直接的抑制作用.作者认为,本研究对用RBX移植后既能成骨及不发生明显免疫排异的机理是一种有意义的探讨,为临床使用提供了免疫学上的依据.
Four groups of female BALB/C mice were implanted into thigh muscle pouch withreconstituted bone xenograft (RBX), bBMP, chemically treated bovine cancellous bone (bCB) andhydroxyapatite(HAP), respectively. The animals were sacrificed on 2,7,14,28 and 42 days and theirspleen cells collected. The supernatant of spleen cell suspension was assessed for IL-2 activity usimg 3H-TdR incorporation technique both in vivo ther RBX boplantaion and in vitro following stimulation bythe homologous antigen. The results revealed that: there was a transient increase in IL-2 level at tWo daysafter RBX implantation; the IL-2 level was seen declining considerably at 7 and 14 days; the CPM for3H-TdR incorporation determined at different time intervals were significanly lowered durin in vitroantigen stimulation, being dependent on the amount of antigen administered. This suggests that RBXimplantation had an inhibitory effect on the T-lymphocyte function and the IL-2 induced CTLLproliferation and it is most ilkely tha RBX also affected IL-2 secretion. The present study power withoutevoking immune rejection, attesting to its suitability for clinical use on the immunological grounds.
