花生中乙草胺残留的免疫亲和柱净化-高效液相色谱分析 被引量:4

摘要 建立了免疫亲和柱净化-高效液相色谱-紫外检测器测定花生中乙草胺含量的方法。样品经过纯甲醇提取浓缩,用含有乙草胺抗体的免疫亲和柱(IAC)净化,甲醇洗脱。以C18柱为分离柱,乙腈-水(体积比75∶25,pH值3)为流动相对洗脱液进行高效液相色谱分离及紫外检测,紫外检测波长为215nm。结果表明:乙草胺IAC柱的吸附容量为0.556μg;仪器的检测限为15μg/L,方法检测限为3μg/L;乙草胺的质量浓度为20~500μg/L时与峰面积呈良好的线性关系,相关系数为0.9969;在5~50μg/kg添加水平下,经过IAC柱浓缩净化提取物,测得回收率在94.06%~102.5%之间,变异系数在2.55%~7.95%之间。 A method was developed for the determination of acetochlor in peanut by high performance liquid chromatography with ultraviolet detection after IAC clean-up. Acetochlor in peanut was extracted with methanol, concentrated and purified by IAC containing antibodies specific for acetochlor, washed the IAC with methanol and quantified by high performance liquid chromatography with C18 column, mobile phase of acetonitrile-water(75:25, by vol,pH=3) and ultraviolet detection with wavelength at 215 nm. The absorb capability of acetochlor IAC was 0.556μg. The limit of detection of instrumental was 15 μg/L and the limit of detection of method was 3 μg/L.The concentration of acetochlor was linear with the area of detected peak ranging from 20 to 500 μg/L and the correlation coefficient was 0.9969. After concentrating and purifying by IAC, the recoveries were from 94.06 to 102.5%, the relative standard deviations were from 2.55 to 7.95% at the spiked levels range from 5 to 50 μg/kg.
