摘要
用ABC免疫组织化学染色法,观察猪卵透明带单克隆抗体(LPD_8、LPC_4),在体外与人、猪卵巢组织的反应。LPD_8和LPC_4使人、猪卵透明带及卵细胞质着色,但不使卵巢的其他结构,如颗粒细胞、卵泡膜细胞及结缔组织着色。这些结果提示:猪卵透明带单克隆抗体对卵巢内分泌功能可能无直接影响,但可通过与卵细胞质结合,抑制卵泡的发育,继而导致卵泡数目减少和内分泌功能降低。作者认为,用本实验提取的两株猪卵透明带单克隆抗体,进行免疫避孕研究前途不大。
Reaction between monoclonal antibodiesto porcine zona pellucida(LPD_8,LPC_4)andhuman and porcine ovaries was studied byABC immunohistochemistry technique invitro.When slices of human and porcineovaries were stained with ABC, not only thezona pellucida but also the ovum cytoplasmwas stained. Other regions of the ovary, suchas granulosa cell,theca cell and connectivetissue,could not be stained. These resultssuggest that the monoclonal antibodies toporcine zona pellucida probably have nodirect effect on the endocrine function ofovary,but they may combine with ovumcytoplasm and. inhibit follicular growth, sothat the number of follicles reduces andendocrine function of the ovary decreases.We do not think that the prospects ofcontinual study on LPD_8, LPC_4, are good.
