摘要
作者用0.01 mol/L柠檬酸提取家兔中性粒细胞溶酶体可溶性成份,经制备性聚丙烯酰胺凝胶电泳,从提取物中纯化出5种肽(NP_1、NP_2、NP_3、NP_4、NP_5),它们的分子量在2500~6000之间,体外杀白色念珠菌试验证实,NP_1、NP_2有高效杀菌活性,NP_3也有较强的杀菌活性。
Five antimicrobial peptides were extrac-ted from the cytoplasmic granules of rabbitneutrophils with 0.01 mol/L citric acid.SDS-PAGE gel densitometric analysis of thecitric acid extract showed that they ac-counted for 34%-42% of the total extractproteins. When subjected to acid urea poly-acrylamide gel electrophoresis, five peptidesmigrated much further to the cathodethan lysozyme. Beginning with the mostcathodal component, they were in turnreferred to as NP_1, NP_2, NP_3, NP_4, andNP_5. They were purified from the citricacid extract by virtue of preparative poly-acrylamide gel electrophoresis.Each of themwas of low molecular weight (2500-6000)asdetermined by SDS-PAGE. Three of the peptides were tested invitro for fungicidal activity. When Candidaalbicans (10~7/ml) were incubated with 12.5μg/ml of NP_1 or NP_2 at 37℃ for 2 h, thedeath rates of the organisms were 96.6%and 93.3% respectively. NP_3 was shown tobe active against Candida albicans althoughless potent than NP_1 and NP_2.
