摘要 固相肽合成(SPPS)技术是目前制备各种模式多肽和天然多肽类似物的最有效方法,但是最终产物的成分往往比较复杂,不经纯化难以用于蛋白质多肽的结构、功能和药理学研究。近年来已出现了多种多肽及蛋白质的分离纯化技术,高效液相色谱法(HPLC)是目前分析纯化合成多肽的主要手段。本文根据固相肽合成方法的原理及产物特点,对高效液相色谱法在分析纯化合成肽中的应用作了较为系统的评述。 Solid phase peptide synthesis technology is the most effective way in preparation of various modes of peptides and natural peptide analog. However, the composition of the final product is often more compfieated, which is difficult for the study in structure, functions and pharmacology of protein and peptide without purification. In recent years there have been a variety of peptide and protein separation and purification technologies, and High Performance Liquid chromatography (HPLC) is the primary means in purification of synthetic peptide. Based on the principles of solid phase peptide synthesis methods and the product features, a more systematic review in the application of high performance liquid chromatography in the analysis and purification of synthetic peptide was made in this paper.
