摘要
目的研究纯化麦冬总皂苷的工艺条件。方法利用AB-8大孔吸附树脂吸附麦冬总皂苷,用乙醇梯度洗脱,确定麦冬总皂苷的吸附容量,考察洗脱条件,确定洗脱参数。结果AB-8大孔吸附树脂吸附容量为53.16 mg/ml,乙醇浓度在80%洗脱麦冬总皂苷效果最佳,回收率为85.49%。结论该方法简单,方便,纯化效果良好,结果满意。
Objective To purify the total saponins in root of Ophiopogon japonicus. Methods We made use of AB - 8 resin column to absorb saponins and alcohol to elnte in different concentrations, confirmed the static absorption capacity of total saponins and studied elution requirement and parameter ascertainment of AB - 8 resin. Results The total saponin absorption amount of AB - 8 resin column reached 53.16 mg/ml. When the alcohol was used for washing buffer, the best concentration of alcohol for eluting saponins was 80% and the recovery rate was 85.49%. Conclusion This method is sensitive and accurate.
