摘要
对从大连小平岛地区海面下10~20m处的海泥样品中分离的5株海洋放线菌进行了生理特征和抗菌活性的研究。抗菌活性实验初步表明,菌株S097,S187和$233具有较好的拮抗革兰氏阳性菌、革兰氏阴性菌和真菌测试菌株的活性,尤其是菌株S233对绿脓杆菌和白色假丝酵母的抑制活性很强。16srDNA序列分析结果表明,5株放线菌(S097,S187,S233,S239,L180)分另0与Streptomyces argenteolus CGMCC4.1693、S.flavofuscus NRRLB-8036、S.variabilis NRRLB-3984T、S.1itmocidini NRRL B-3635和S.sulphureus NRRL B-1627T显示出最高的序列同源性(99%),这是这些菌种首次报道在大连地区的海泥样品中得到分离。利用Ⅰ型聚酮合成酶(PKSI)兼并引物从菌株S187中扩增出了PKSI片段,揭示了该菌株生产Ⅰ型聚酮类化合物的潜在能力。本文的研究结果为进一步开发利用大连地区海泥中的海洋放线菌资源奠定了基础。
The physiological features and antimicrobial activities of five marine-derived actinomycete strains isolated from the sediment samples from Dalian sea area were studied. Isolates S097, S187 and S233 showed broad antagonistic activities to various gram positive, gram negative bacteria and .fungal test strains, and isolate S233 displayed strongest activities against Candida albicans and Pseudomonas aeruginosa. Analysis of the 16s rDNA sequences revealed that the five isolates (S097, S187, S233, S239, L180) showed the highest homology (99%) to Streptomyces argenteolus CGMCC 4. 1693, S. ffavofuscus NRRL B -8036, S. variabilis NRRL B-3984T, S.litmocidini NRRL B-3635 and S. sulphureus NRRL B-1627T, respectively. This is the first report that these species were isolated in the marine sediment samples in Dalian sea area. Furthermore, polyketide synthetase Ⅰ (PKSI) fragment was amplified from isolate S187 using the degenerate primers, implying the potentiality of this strain in the production of type Ⅰ polyketides. These studies may provide the basis for further exploration of marine-derived actinomycetes in Dalian marine sediments.
