摘要
目的:建立HPLC梯度洗脱法测定天山花楸叶中芦丁含量的方法。方法:采用ZORBAX SB-C18(4.6mm×15mm,5μm)色谱柱,甲醇-0.02%磷酸溶液为流动相(梯度洗脱),流速为1.0ml·min^-1,检测波长为326nm。结果:芦丁线性范围为0.2104-1.8936μg/ml,叶中芦丁含量0.4829mg/100ml(RSD%=0.48%,n=3),平均回收率为100.4%。结论:该方法经济、简便、快速、准确、可靠,可用于该药材的质量控制。
Objecthe: To establish the method of Rutin determination in Sorbus Tiansehaniea. Methods: ZORBAX SB - C18 (4.6mm ×15mm, 5μm) column of High Performance Liquid Chromatography ( HPLC ) was adopted in the Rutin determination. The MeOH-0.02% H3PO3 was used as mobile phase. The flow rate was 1.0ml·min^-1. The detective wavelength was 326nm. Results: The linear range of Rutin was 0.2104 - 1.893μg/ml. Rutin in the leaf of S. Tianschanica was 0.4829mg/100ml (RSD% = 0.48%, n = 3), and the average recovery- rate was 100.4%. Conclusion: The method is simple and sensifive, and can determine Rutin content quickly and correctly. It is suitable for the analysis of Rutin in S. Tiansehanica.
