摘要
目的探讨谷氨酰胺(GLN)二肽全肠外营养对急性放射性肠炎大鼠脾脏的营养作用。方法应用19只SD大鼠行全腹部放射后,实验动物随机分为三组:常规肠外营养组(STD组,n=6),大鼠输入常规肠外营养液;谷氨酰胺组(GLN组,n=7),大鼠输入含谷氨酰胺的静脉营养液;正常组(n=6)为未经腹部照射的正常大鼠,连续7d肠外营养。实验结束前经颈静脉注入5-溴脱氧尿苷(BrdU),光镜下观察脾脏的形态学变化及脾脏BrdU阳性细胞,电镜下观察脾脏淋巴细胞超微结构的变化。结果GLN组脾脏白髓内淋巴细胞增多,脾脏BrdU阳性细胞数多于STD组,电镜下观察GLN组可见明显的淋巴细胞激活征象,表现为核染色质凝集,异染色质减少,胞质内线粒体和粗面内质网增多。结论GLN二肽可促使受辐射性损伤后脾脏内淋巴细胞的增殖,并保护淋巴细胞的结构。
Objective To study the nutrient effect of glutamine-enriched parenteral nutrition on rats' spleen of acute radiation enteritis. Methods Nineteen SD rats were used, which received whole abdominal radiation. The animals were randomly divided into three groups. Standard total parenteral nutrition (STD group, n=6) and glutamine-enriched total parenteral nutrition (GLN group, n=7) was used from intravenous infusion continuously for 7 days, and Normal nonirradiated animals (n=6). The samples of spleen from these animals were taken after intravenous injection 5-bromodexyridine (Brdu) via internal vena for 1 hour. The histological changes of spleens and bromodexyridine labelled cells were studied by using light microscopy. The microstructure changes of spleen lymphocytes were studied by using transmission electron microscopy (TEM). Results The lymphocytes of white pulp, periarterial lymphatic sheath and spleenic nodule of GLN group increased compared with those of STD group. The BrdU labelled cells of the spleenie lymphocytes in GLN group were significandy more than that of STD group. Under TEM, the lymphocytes of spleen of GLN group revealed that the ehromatin agglutinated and the heteroehromatin decreased, the mitochondria and the rough endoplasmic reticulum increased, and the mitochondria had a little swollen compared with those of STD group. Conelusion Glutamine-enriched parenteral nutrition can improve the spleen lymphocytes proliferating and maintain the spleenie structures after abdominal radiation.
