摘要
目的检测灯盏花素对实验性糖尿病大鼠视网膜血管内皮生长因子(VEGF)表达的影响,探讨灯盏花抑制糖尿病视网膜病变的可能机制.方法采用链尿佐菌素诱导大鼠糖尿病模型,将动物随机分为糖尿病组(10只×2)和注射用灯盏花素治疗组(10只×2),另取等量正常大鼠做对照组.治疗组在出现糖尿病表现后按每日20mg/kg腹腔注射灯盏花素溶液,糖尿病组和对照组同期注射等量生理盐水.注射后2、8周取左眼视网膜行逆转录聚合酶链反应(RT-PCR),右眼行免疫组化分别观察VEGFmRNA和蛋白的变化.结果2周糖尿病组VEGFmRNA较对照组明显增加(P<0.05),蛋白表达无明显变化;灯盏花素治疗组VEGF较糖尿病组无明显变化.8周糖尿病组VEGFmRNA和蛋白表达均较2周时明显增加(P<0.05),而治疗组两者较糖尿病组明显减少(P<0.05).结论灯盏花素明显抑制糖尿病大鼠视网膜VEGF表达,提示灯盏花素可能通过降低糖尿病大鼠视网膜VEGF而缓解其糖尿病视网膜病变.
Objective To observe the effects and significance of Herba Erigerontis the expression of vascular endothelial growth factors (VEGF) in the retinas of diabetic rats. Methods Diabetic rats by streptozotocin were randomly divided into diabetes group (10 × 2) and Herba Erigerontis treatment group (10 × 2). Another 20 normal rats were recruited as normal control group. The Herba Erigerontis reatment groupwas injected Herba Erigerontis ( 20 mg/kg) while diabetes groups and normal control group were injected saline solution into peritoneal cavity once every day. 2 weeks or 8 weeks later, 10 rats in each group were killed and the expression of VEGF in the retinas were detected by RT-PCR and immunohisochemistry. Results In 2 weeks, the expression of VEGF mRNA in diabetic group increased and it were significantly higher than that of streptozotocin treatment group (P 〈 0.05) , but the level of protein did not have different. In 8 weeks, the expression of VEGF mRNA and protein in diabetic group further increased and they were remarkably higher than that of Herba Erigerontis treatment group (P 〈 0.05). Conclusion Herba Erigerontis injected intraperitoneally decreases the expression of VEGF in retinas of diabetic rats so that partly suppresses the development of diabetic retinopathy.
